An Alba-domain protein required for proteome remodelling during trypanosome differentiation and host transition

چکیده
The transition between hosts is a challenge for digenetic parasites as it unpredictable. For Trypanosoma brucei subspecies, which are disseminated by tsetse flies, adaptation to the new host requires differentiation of stumpy forms picked up from mammals procyclic in fly midgut. Here we show that Alba-domain protein Alba3 not essential mammalian slender forms, nor required culture or mice. It crucial, however, development T . during transition. While steady state levels mRNAs differentiating cells barely affected loss Alba3, there major repercussions proteome. Mechanistically, aids rapidly releasing translational repression and stimulating polysome formation. In its absence, fail remodel their proteome appropriately, lack components mitochondrial respiratory chain reduced infection tsetse. Interestingly, closely related Alba4 functionally redundant but cannot compensate form. We postulate proteins play similar roles regulating translation other protozoan parasites, particular life-cycle transitions.

ALBA proteins are stage regulated during trypanosome development in the tsetse fly and participate in differentiation
The protozoan parasite Trypanosoma brucei is responsible for sleeping sickness and alternates between mammal and tsetse fly hosts, where it has to adapt to different environments. We investigated the role of two members of the ALBA family, which encodes hypothetical RNA-binding proteins conserved in most eukaryotes. We show that ALBA3/4 proteins colocalize with the DHH1 RNA-binding protein and ...
متن کاملThe Trypanosome Pumilio Domain Protein PUF5
PUF proteins are a conserved family of RNA binding proteins found in all eukaryotes examined so far. This study focussed on PUF5, one of 11 PUF family members encoded in the Trypanosoma brucei genome. Native PUF5 is present at less than 50000 molecules per cell in both bloodstream and procyclic form trypanosomes. C-terminally myc-tagged PUF5 was mainly found in the cytoplasm and could be cross-...
متن کاملBrg1-dependent chromatin remodelling is not essentially required during oligodendroglial differentiation.
Myelinating Schwann cells in the vertebrate peripheral nervous system rely on Brg1 (Smarca4) for terminal differentiation. Brg1 serves as central ATP-hydrolyzing subunit of the chromatin remodelling BAF complexes and is recruited during myelination as part of these complexes by the transcription factor Sox10 in Schwann cells. Here, we analyzed the role of Brg1 during development of myelinating ...
متن کاملloco encodes an RGS protein required for Drosophila glial differentiation.
In Drosophila, glial cell development depends on the gene glial cells missing (gcm). gcm activates the expression of other transcription factors such as pointed and repo, which control subsequent glial differentiation. In order to better understand glial cell differentiation, we have screened for genes whose expression in glial cells depends on the activity of pointed. Using an enhancer trap ap...
